Detection of mutagens by automated microbiology system-2(MS-2)

Abstract

|
The mutagenicity of N-methyl ?N¡¯-nitro ?N¡¯-nitrosoguanidine(MNNG) by both Ames plate method and a modified Ames assay adapted to the automated MS-2 Microbiology System was investigated.
MNNG was strongly active in the reversion of the base-pair substitution mutation in S. typhimurium TA1535 and TA100, but did not show any mutagenic potential in TA 98 and TA102 of Ames plate assay.
The more MNNG was administered(from 51ug/plate to 100, ug/plate), the less the revertants of TA1535 and TA 100 could be found.
The mutagenic results of MNNG on TA100 were able to be obtained much faster than those of MNNG on TA1535 by MS-2.
MS-2 exhibited the comparable reversion of TA1535 and TA100 by MNNG with a minimal requirement for materials and labor.
